The free version is very limited, though, and the screenshot feature saves to BMP which means it’s only slightly more modern than having someone draw it on parchment with a quill. You can tell Fraps has been around for a while: how many screen recorder apps still list Windows XP and 2003 in their compatibility list? This is an old app – the most recent release came out seven years ago – but its age is actually a bonus if you’re running older versions of Windows or need a screen recording app with very modest hardware requirements. One thing that’s different about Screen Grabber Premium is the pricing: if you don’t shell out for the lifetime licence, your money only pays for a year of using the app. The interface is pretty minimalist but it’s clear enough and straightforward to use. The built-in editor enables you to add text, lines, call-outs, and other on-screen items, which is useful if you want to create a tutorial rather than just capture your gameplay, and the app can also record audio from anything that’s playing on your PC or Mac. Screen Grabber Premium enables you to record full screen, a selected area, just your webcam, or even just a small area around your mouse, and it outputs in all the key formats, including MP4. Running the old msdbg on the memory dump reveals the following, indicating to my eye that peauth.sys for some reason fainted during bootup:ĮRROR_CODE: (NTSTATUS) 0xc000001d - Illegal Instruction An attempt was made to execute an illegal instruction. I couldnt be happier with the motherboard and Core i5 650 if this issue got fixed. Please note, I'm not overclocking anything. The PC will now function perfectly and indeed perform very well. I then reboot, enter BIOS again end enable the onboard NIC. The PC boots perfectly with no network ofc. I can work aound this by first entering BIOS and disable the build-in NIC. Upgrading from the december 2009 BIOS actually was an improvement. Im using the March 2010 BIOS, which at present seems to have been withdrawn from the Intel site. I think I ran into an incompalibility between the 82578DC NIC and Microsofts bothersome build-in license software. ![]() I don't have any more to build for a bit, and in the future I will probably be using the DP55WB board if I am not gong to use the integrated video, but for me, this explains all 3 systems and my confidence in this board has been restored. NOW I am not claiming this would fix it for everyone, but I am pretty sure this is what I was running into and it started working form me when I did other things, but in reality, it was the hard power cycle after the bios upgrade that fixed it every time. After that, it started working and it hasn't failed yet. IT STARTED WORKING when I gave it the hard power cycle and made sure power was cut to the system board (I think I did this with the network cable unplugged). After the upgrade - NETWORK FAILED! I then tried rebooting with and without the cable plugged in, but it still didn't fix it. I intentionally left the network cable plugged in while performing the bios upgrade. I then loaded the additional drivers one by one, and between each driver I did all 3 (reboot, shutdown and turn on, shut down-hard power cycle-turn on) and it worked every time. I tried to get it to fail, restarting, shut down and power on, shut down, pull the plug, let the power dissipate and then plug back in and turn on, AND IT CONTINUED TO WORK. ![]() I then installed chipset and network drivers and it worked. So on my 2nd system, I still left the network cable unplugged. I ended up using msconfig to force default video mode (see previous post) and then reenable the regular video mode and it started working and hasn't quit yet. ![]() I tried to disable it and reenable but still didn't work. I continued to load the rest of the drivers and when I was done the network card quit working again. I loaded the chipset driver first, then the network and the network showed a cable was unplugged, I plugged it in and it was working. On the first one, I made sure I installed Windows with the network cable unplugged. ANOTHER UPDATE- I have built 2 more of these machines. ![]() One of the simplest types of epistasis is that which occurs between mutations within a single protein. If the same is true for other proteins and viruses, this work could lead to a deeper understanding of the constraints that govern evolution at the molecular level.Įpistasis can play a key role in evolution, such as by constraining accessible evolutionary pathways ( Weinreich et al., 2005 Kryazhimskiy et al., 2011) and increasing the role of contingency in adaptation ( Blount et al., 2008 Bridgham et al., 2009). Overall, these results show that interactions among mutations constrain the evolution of the influenza nucleoprotein in a fashion that can be largely understood in terms of protein stability. Intriguingly, the constrained mutations helped the virus overcome one form of human immunity to influenza, suggesting that interactions between mutations might limit the rate at which viruses evolve to evade the immune system. It may, therefore, be possible to identify enabling mutations based on their effects on thermal stability. The constrained mutations decreased the stability of the nucleoprotein at high temperatures, while the enabling mutations counteracted this effect. They found that several mutations greatly reduced the fitness of the 1968 virus when introduced on their own, which strongly suggests that these ‘constrained mutations’ became part of the virus’s genetic makeup as a result of interactions with ‘enabling’ mutations. introduced each one individually into the 1968 nucleoprotein. To test whether all of these mutations could have been tolerated by the 1968 virus, Gong et al. ![]() Between 19, the nucleoprotein-which acts as a scaffold for the replication of genetic material-in the human H3N2 influenza virus underwent a series of 39 mutations. examined whether such interactions have indeed constrained evolution of the influenza virus. This type of mutational interaction would constrain the evolution of the virus, since its capacity to take advantage of the second mutation depends on the first mutation having already occurred. Similarly, during the evolution of a virus, a mutation that helps the virus evade the human immune system might only be tolerated if the virus has acquired another mutation beforehand. In this example, the D must be changed to an E before the W is changed to a G, because GORD is not a valid word. This phenomenon is similar to the game in which one word is converted to another word, one letter at a time, subject to the rule that all the intermediate steps are also valid words: for example, the word WORD can be converted to the word GENE as follows: WORD→WORE→GORE→GONE→GENE. Our results paint a coherent portrait of epistasis during nucleoprotein evolution, with stabilizing mutations permitting otherwise inaccessible destabilizing mutations which are sometimes of adaptive value.ĭuring evolution, the effect of one mutation on a protein can depend on whether another mutation is also present. The constrained mutations occurred at sites enriched in T-cell epitopes, suggesting they promote viral immune escape. These mutations were destabilizing, and were preceded or accompanied by stabilizing mutations that alleviated their adverse effects. Several mutations were deleterious to the parent despite becoming fixed during evolution without negative impact. To test whether epistasis similarly constrains actual protein evolution, we created all intermediates along a 39-mutation evolutionary trajectory of influenza nucleoprotein, and also introduced each mutation individually into the parent. In this analogy, epistasis constrains evolution, with some mutations tolerated only after the occurrence of others. John Maynard Smith compared protein evolution to the game where one word is converted into another a single letter at a time, with the constraint that all intermediates are words: WORD→WORE→GORE→GONE→GENE. ![]() Conversely, contemporary interiors adhere to a more minimalist, practical design aesthetic that uses space better and highlights natural light.īecause of this contrast, one of the finest ways to update a formal dining room is to experiment with space and lighting. The colorful conference room where she and Dickson chatted after her tour informed the look of the fictional Mattel’s inner sanctum. Gerwig would go on to write and direct “Barbie,” and her brand immersion left some visual imprints. Just like the Design Center exhibition, the film could bring together Barbies from across the decades: Robbie could be one of many. “Barbie” could be a meta-comedy that moved between the idealized, plastic realm of the dolls and the human world of the company that had created them. “We’ve had a Barbie President, I see.” A Mattel employee proudly replied that Barbie had been to space before most earthly women had a credit card.Īs Gerwig peppered Dickson with questions, a concept began to take shape. Gerwig was shown a display called the Barbie Wall of Fame, whose entryway was emblazoned with the brand’s mission statement: “ to inspire & nurture the limitless potential in every girl.” Framed photographs captured Barbie posing with everyone from Diahann Carroll to Dean Martin, but Gerwig was most struck by a series of shots of historical models, including an all-Barbie Presidential ticket, from 1992. At one point, Dickson called her when she was mixing “Little Women” in New York. She saw in Gerwig’s filmography the right combination of intelligence and heart: “You watch something like ‘Little Women,’ and the dialogue is very, very clever-it’s talking about some big things-but it’s also extremely emotional.” The project wasn’t an obvious fit for someone whose screenplays included the subtle dramas “ Lady Bird” and “ Frances Ha,” and Gerwig wavered for more than a year. She’d signed up for a Barbie movie, and had approached Gerwig about writing the script. The actress Margot Robbie, who had toured the center in 2018, wanted to continue playing. And, if you don’t walk out drinking the Kool-Aid, then it was a great playdate, but maybe we don’t continue playing.” “I have met with some of the greatest artists, truly, in the world. . . . “The brand immersion is the everything moment,” Dickson told me. ![]() The center even boasts a giant rendering of Castle Grayskull, the fearsome ancestral home of He-Man. It’s important to keep your emergency fund and checking account separate. Savings Account: Online Bank or Credit Union Quick answer: in a savings account at an online bank or a credit union. It’s great if you can eventually store six months’ worth of expenses in an account that’s easily accessible. If you’ve already set up automated transfers to your emergency fund and you want to quantify your goal, figure out the total of your household’s monthly expenses. It’s better to save first rather than waiting to see if you have any money left over. He recommends setting up an automatic transfer into the account that holds your emergency fund every paycheck or once a month. “And then, when you’re in that habit, you really can move toward the goals that are really going to change your financial future.”Ĭlark likes automated savings. “It’s all about building habits, whether it’s with your health or your wallet, that you do it one step at a time,” Clark says. Then increase the amount you’re contributing over time. Just get in the habit of contributing to your savings account. Don’t feel pressured to meet a quota or a timeline. If you don’t have any savings, take $10 from your next paycheck and put it into a savings account. How much money should you keep in an emergency fund? It’s a fair question, but if you’re just getting started as a saver, focusing on the number may not be helpful.Ĭlark says that your first priority as a saver is to start somewhere. How Much Money Do I Need in My Emergency Fund? Worse, many Americans have to pay for them with high-interest credit cards or by borrowing money.
